Peptide synthesis – synthesis and membrane interaction studies of internalising peptides of viral origin
Solid phase peptide synthesis (SPPS) with Fmoc/tBu strategy, manual technique, will be applied to synthesise labelled peptides of viral origin. The „pupil” is going to learn the general technique of SPPS, the applicability of orthogonal protecting groups, monitoring of coupling, cleaving and purification. After the completion of the program she will be able to synthesise oligopeptides independently. During the training, she will have opportunity to get familiar with automatic and pin peptide synthesis methods as well.
Cell targeting peptides of viral origin will be prepared and their internalisation ability will be studied. The interaction with cell membrane is an important part in understanding the structure – activity relationships. Therefore the synthetic peptides will be studied on modell membrane systems with circular and linear dichroism, as well as infrared spectroscopies.
During the training period experience can be gained both in the field of solid phase peptide synthesis and spectroscopic characterisation.
